The Workable Business Blogging Plan - Blogging Tips

I wanted to put together a plan to help people who are wanting to build a business using blogging. There are a lot of the workable business blogging planpeople telling you that you can make money online blogging and they are not lying, you can make good money from blogging but it isn?t particularly easy especially if you do not have a plan or follow a system.

Writing blog posts every now and then will not build you a business; waiting for the advert breaks in your favourite TV shows to write a quick post simply will not create the content that you need to build a blog big enough to make money.

In a nut shell, you need a plan, if you don?t have a plan you do not have a linear train of thought and nothing to work towards. You will just be ambling around bouncing from one job to another and getting nowhere.

It sounds clich?d but this saying really does sum it up? ?Fail to plan, plan to fail? simple.

For new bloggers just starting out I wanted to think up a plan that could greatly help them to become more successful quickly. Use this as a guide, do the bits you want to do and forget those parts you don?t. This guide is an example for you to work with and create your own plan. Of course you can follow it as it is. Whatever you do, you need to have a plan or schedule to work with to even come close to being successful.

What follows is my plan for starting a blog and building it quickly up to a point where it is growing thanks to the help of others leaving you to focus on other tasks.

1.?????? Register a Domain and Get Hosting.

Obviously you will need a website to get started, you might already have your website set up, if so then this part is complete. Both domain registration and hosting can be done using Hostgator. Without hosting or a domain name you won?t have an online business. For help setting up your website click here.

2.?????? Install WordPress

Next you need to install WordPress, this is easy to do in the CPanel area you get when you buy hosting. If you do not yet have hosting and think creating your blog is hard we can do it for you for free here. You can use a free WordPress Theme or you could invest in a premium Theme like those found at Woo Themes. Premium Themes can give you more functionality and scope than the free ones.

3.?????? Add An Email Opt In Form To Your Site

From day one you should start to build an email list. Create an Aweber?account and build an email capture form to install on your website offering a monthly newsletter. There are different kinds of email newsletters, some people write a post good enough to be on the blog that they send to their email list as a subscribers only bonus post, others send a shorter email with advice and some people send a weekly roundup.

The easiest email newsletter to send is the weekly roundup; if you are publishing a new post everyday you have 7 new posts to remind people about. With so much content being created this type of email gives you plenty to add in the email without having to write new content.

When you build an email list you create a list of people to promote future posts, products and promotions to. You can send traffic back to your blog which will be noticed by the search engines, they love sites that have returning traffic. Building an email list is one of the most important things you can do when you start an online business.

4.?????? Create Twitter, Facebook, YouTube & Google+ Accounts?social logos

Get these in place when you start your blogging business and use them to build a following to send?traffic back to your

blog. Every time you write and publish a new post share it on these networks. When you create a video to share on your blog, upload it to YouTube and host it there, videos in YouTube can be taken and use by other sites as well as appear in the Google search results. This is great for getting free traffic.

A lot of the social media work can be controlled and automated by using software like TweetAdder for Twitter or Hootsuite which is ideal for most if not all of your social media networks, all of them accessible from one dashboard without having to log in to each one separately every time you want to share your latest post.

5.?????? Create 1 New Quality Post Per Day For 3 Months.

This will be the hardest part and will take some time getting used to. You will need discipline and motivation. You now need to write at least one quality post to be published per day. These posts need to be 800 words or more and very helpful.

They need to be of the best quality possible, Google and readers love quality, your readers need to leave your site feeling that they have learned something new and valuable.

You can create video posts and podcasts but they would still need to have plenty of good written content with it. The idea is to give the readers plenty of useful information. To keep costs down you need to write and create your content yourself. You can outsource it if you want but that is your choice.

You may decide to publish fewer posts per week but that will create less powerful results. You need to be consistent, stick to getting a quality new post published every other day if you believe everyday is too hard going for you.

3 months of solid work and you should start to see movement in the search engines and get traffic to your site, I say should, for some people it might be more or less depending on what other tasks you perform during that 3 months. If it doesn?t happen in 3 months, do not despair, it will happen just keep at it. This is a business and you should be thinking long term not short term anyway.

6.?????? Write 2 Guest Posts Per Week.

Write 2 guest posts or more each week and get them published on other sites to create backlinks to your site as well as start to create a recognised name for your site. The better your guest posts are then you stand a better chance of having it accepted on sites with higher page rank. Getting a post published on a site with a good page rank will be better for you.

Find sites asking for guest posts and send them an email offering your post to them. Every site requires a new unique post for their site only. There is a science to writing a good guest post and you need to make sure that you write the best posts possible. Zac Johnson says that ?your guest posts should be better than those on your own site?

7.?????? Join Guest Blogging Sites

There are networks where you can join and submit posts to be used as guest posts; you will also find people who have submitted their own posts and articles looking for sites to publish them. You can find some potential good writers in these networks and this is in preparation for a following stage. Some guest posting sites are GuestBlogIt.com, MyBlogGuest.com, Guestr.com and BlogSynergy.com

8.?????? Start Commenting On Blogs & Forums

This is a must do job, in the first 3 months you should be commenting on other same niche sites and building up a network of friends and more importantly new readers. You should not be just adding crap useless comments but you should in fact be discussing the post you have read and making friends.

Offer opinions, tips, advice and personal stories on the post topic, you need to interact with other commentators and the post writer. You are being seen and creating a healthy trust as well as creating a network along with hijacking some of the big sites traffic for free.

Many of these comments can get you links back to your site which can help but it is really all about the relationship building and free traffic. To be successful at blog commenting you need to read the post, otherwise you will not know what to write about and will look like you are leaving comments for spam reasons.

Create a spreadsheet of the best blogs in your niche and visit them as often as possible. John Chow (who now makes $40,000 a month from blogging) says that in his early days he would leave comments on at least 3 different blogs each and every day. If you think you do not have the time you could do this task while having a tea break or during lunch.

Failing that you could hire a close friend or someone in your family like a child or nephew to do this job for you as they sit and watch TV. Offer them ?-$10 a week to place a set amount of comments each week as long as they read the posts and leave quality comments that sound like you. It would be wise to have them record the posts they comment on each week and that way you can check that they are not simply leaving ? ?nice post, I have bookmarked and will return again?

9.?????? Create A Write For Us Page

Create a write for us page outlining exactly what it is you want and what you will accept. Try to wean out as much wheat from the chaff as possible from the beginning. Include a contact us form at the bottom of the page for writers to contact you and submit their posts.

By doing this you will soon be sent posts that you can add to your blog giving you free content and start taking some of the pressure off of your writing schedule. You might also find potential good writers who could help you grow your blog when it is ready for stage 10.

10.?? Invite Your Favourite Writers To Write For You

Once your site has grown and has a good number of monthly visitors and readers you can invite some of your favourite writers or guest post suppliers to write for you on a more permanent arrangement. Create an author account within your WordPress site giving them their own login details so they can add and schedule their posts to go live without your interaction.

You can set the rules so that every post has several internal links sending readers to related posts on your site and that they can then add one link back to theirs, you could suggest that each new posts has to be scheduled for a certain amount of time giving you the option to check that they are up to your standard so you can delete any if required and add affiliate links to the article.

Take a look at this site and you will see that there are several writers for the it even though it is owned by Zac Johnson. You could do the same, if your writers were happy to post 1 new article a week then you need a team of only 7 writers. If you wrote one post yourself then all you will need is 6 writers.

How hard can it be to create a team of great writers all willing to write for you? Remember they will get extra exposure helping to build up their names and brands also their sites receive free traffic and links. They get back just as much as they put in. Both parties win from this arrangement.

You could have a team of 14 writers all adding just one post every other week or have a team as well as submitting the one off guest posts that you will still receive. I have had several posts published on sites even though I wasn?t a member of the sites main writing team.

Everyone who has a post published on your site whether they are a guest blogger or one of your new writing team will share their posts as well as some of the others on their social media networks. This can create an influx of traffic to your site from their large lists of followers.

What really matters is that your site is getting quality content that is being published each day. This will please Google and it will please your readers and it will really please you in the long run.

11.?? Let The Writers Create The Content & You Focus On The Business

Now that you have a team of writers adding content to your site for you, you can slow down on the writing schedule and now focus on the other aspects of building your online business. You can go looking for those great affiliate offers that can generate you money or focus in finding people to advertise their services on your site earning you a monthly income.

There are a lot of jobs that need doing when you are building an online business, from WordPress maintenance to traffic generation. You could focus your writing skills to creating free reports and eBooks to give away for traffic generation, email list building and relationship building. These reports could have affiliate links within them and become another source of affiliate commissions.

You could write more guest posts and have them submitted to other new sites creating new links and more free traffic sending readers to your now well stocked and live blog. You could use your time creating great ?How to? video tutorials or create a series of videos and start a monthly membership course or product.

Eventually you will hit the tipping point where your site will grow on its own and you will benefit from all of your hard work. It might take 3 months or it might be longer, what is important is that you do not expect to be successful after just 3 weeks and give up. In the online and internet marketing world dreams have been sold and promises of fast riches were made so people now expect it to happen overnight.

The truth is this, if someone makes a lot of money fast they are the exception to the rule and not the norm. This is a business and you are building it, we know off line businesses require time to build and we should approach the online business in the same way.

We should mental allow ourselves at least a year or two before we expect to see results, if they come sooner then that is great but if we do not think in terms of a year or more we would become disheartened too quickly and give up, this is not what we want you to do.

So there you have it, a plan or guide depending how you want to see it of a way to get focused and build your blog business quickly. Scheduling time to write and time to do the blog comments along with all of your other tasks will help you to keep more focused and achieve your goals.

Create your own plan, get serious about your work, do each task daily with discipline and make success happen.

New home sales surged in January, rising almost 16% from December in another sign of an improving housing market.
Sales of new single-family homes in January came in at a seasonally adjusted annual rate of 437,000, the government said Tuesday.
The results bested consensus estimates of 381,000.
new home construction picThe pace of sales was almost 29% above the January 2012 estimate of 339,000, and almost 16% above the revised December rate.
The strong showing in January follows an uptick in home builder sentiment and housing starts in recent months that hadn?t been matched with an increase in sales. ?Now, we?re seeing sales pick up,? says Patrick Newport, economist with IHS Global Insight.
The homebuilding industry has been starved for good news for years. Last year?s new home sales totaled 367,000, making 2012 the third lowest year on record for new home sales. They hit record lows in 2011.
IHS Global Insight expects new home sales to rise to 464,000 this year. IHS doesn?t expect new home sales to return to more normal levels, above 800,000 a year, until 2015.
Increased new home sales are a strong indicator of increasing consumer confidence in the overall U.S. housing market, says John Tashjian, principal, Centurion Real Estate Partners.
As buyers absorb existing inventory, builders will ramp up construction starts, which is also give the economy a ?much needed shot in the arm,? Tashjian says.

Cocooning: It?s Back and Thanks to Tech, It?s Bigger

Cocooning is undergoing a metamorphosis: Call it super-cocooning.

Thanks to always-on wireless Internet connectivity and bigger, better TVs that reproduce pixel-perfect high-definition video, cocooning is entering a new evolutionary stage. Consumers are staying home more, watching movies delivered via cable, satellite, Internet or disc, eating in and transforming their apartments and houses into a shelter from the daily social storm.

Fam in KitchThis new level of super-cocooning is affecting Hollywood, professional sports and restaurants across the U.S. ?Everybody is nervous, really nervous,? says trend forecaster Faith Popcorn, who coined the term ?cocooning? in 1981. ?I think we are looking for protection. Almost like the Jetsons, we want to walk around in a little bubble. We are moving toward that.?

Cocooning is not a new behavior. Born out of a mix of fear and fun, it became a trend identified with Cold War unease that led to stay-at-home entertainment such as the first home video game systems, rec rooms and the adoption of home swimming pools and trampolines.

After the 9/11 terrorist attacks, a refocus on cocooning occurred. Homeowners lined their nests with media rooms and remodeled kitchens meant for entertaining. And in the last 12 months, with the July 20 movie theater shooting in Aurora, Colo., and the Dec. 14 school shootings in Newtown, Conn., many have a heightened sense of unease. ?We don?t feel too safe, and people are getting more and more nervous about being vulnerable,? Popcorn says. ?Cocooning is going strong in 2013.?

By the numbers
An indication of super-cocooning comes from a recent JPMorgan Chase analysis of credit card spending. Consumers with Chase Freedom credit cards spent significantly more (65%) on electronics such as TVs and tablets during the last three months of 2012 than during the same period the year before, the firm found.

Overall, consumers spent 2% more during the fourth quarter of 2012 than a year before, but spent less on hotels (-21%), car rentals (-26%), restaurants (-16%) and tolls (-8%). ?It does appear that consumers are staying closer to home,? says Phil Christian, general manager for Chase Freedom.

That trend is buttressed by the slowed growth in travel and tourism spending, from about 5% growth in the first three months of 2012 to 2.2% and 0.6% in the second and third quarters, reported by the Bureau of Economic Analysis in December.

On the plus side, movie theaters set a box office record of $10.8 billion in 2012. but overall attendance remained flat, according to Nielsen.

That?s in part because Hollywood is increasingly catering to consumers by getting films from theaters into homes more quickly via on-demand or pay-TV services. Among those who stay close to home, a subset of about 7% of U.S. homes with Internet access are inhabited by ?heavy home entertainment cocooners,? says consulting and research firm Frank N. Magid Associates. They spend nearly $300 each month on pay TV, Internet service, video games, on-demand video, music, books, newspapers and magazines, says Magid.

These heavy cocooners are an affluent, racially diverse group: More than one-third (35%) make $75,000 or more annually. Whites make up 57%, Hispanics 22%, blacks 14% and Asians 7%, according to Magid. The firm conducted the nationally representative survey of 2,540 digital consumers in March 2012.

Even a large portion of digital consumers in the $35,000-$50,000 annual-income bracket identified themselves as heavy cocooners.

?The emerging cocoon of home entertainment is being led by a new, technologically sophisticated and more culturally diverse American consumer,? says Andrew Hare, senior analyst for Magid.

The cost of cocooning
While pay-TV bills have risen about 6% annually, The NPD Group says, more homes are opting for higher-cost packages. About 23% of homes pay more than $100 monthly for cable-delivered pay TV, up from 19% in 2008, Magid found. Homes paying more than $100 for satellite pay-TV service rose to 14% from 10%.

But in other ways, the price of becoming a super-cocooner is falling. As the average price for an HDTV has plummeted, now about 88% of homes have one, according to the Consumer Electronics Association. And bigger TVs, those larger than 55 inches, cost on average $1,400, about 10% less than a year ago, The NPD Group says.

Consumers bought slightly more TVs last year than in 2011, with many upgrading to bigger displays, says NPD analyst Ben Arnold. ?The move toward big screen is part of that (cocooning) story. You?ve got tons of content options. You?ve got TVs that connect directly to the Internet, so you don?t even have to get a DVD; you can call up Netflix or Amazon video services directly on your TV.?

There?s more HD content available, too, he says. ?There are a lot of reasons to stay home and either watch movies or sports on TV. Picture quality has become better and better. Actually being able to see the event or see the game, one might argue that it?s a better experience than in person.?

Among recent TV buyers is Kornel Lelea of Hawthorne, Calif., who bought a new 70-inch Sharp HDTV before his annual Super Bowl party. Three other screens also had the game on, but the new set was the star attraction. ?The technology is so much better. It has 3-D capability, the color saturation,? he says. ?It is just beautiful.?

The 46-year-old L.A. housing inspector watches a lot of sports, movies, as well as the Discovery Channel. ?With the (new) 70-inch now, it?s better than a movie theater,? he says.

And it?s safer, even for a guy who?s 6 foot, 2 inches. ?I?m a big Dodgers fan, but the last time I was at a game someone was actually trying to get stupid with me,? he says. ?I?m a fan, but I?m not going to lose my career or my life over a game, either, you know.?

Recent assaults at sports events have caught the attention of the public and fueled cocooning. A post-game stabbing occurred at the NFC Championship game in Atlanta last month. And in 2011, national attention was turned on Los Angeles after a San Francisco Giants fan was beaten at Dodger Stadium. The National Football League in 2008 enacted a Fan Code of Conduct; last season it toughened the rule by requiring fans kicked out of a stadium to take an online behavior-management course before returning.

While convenience, cost and quality of home theater were the biggest factors cited for staying at home, security was a concern for several others who responded to USA TODAY about the subject on Twitter and Facebook. ?Why leave the comfort of my lazy boy (sic) when I can see/watch a HD football game?? wrote Nathan Tameling.

Said Dave Majewski of Columbus, Ohio, ?It?s cheaper and more comfortable and safer.?

That is a growing consumer sentiment, says Tom Campbell of retailer Video & Audio Center in Los Angeles. He was surprised at the rate at which consumers began snapping up new $17,000, 84-inch Ultra HD televisions after LG Electronics began shipping them in late October.

?We called some of them back to ask, ?Why did you buy it?? We found out that with the ever-increasing violence at sports events people are becoming concerned about their safety,? said Campbell, who called several dozen customers. The three-store chain has sold more than 100 Ultra HD displays.

Other retailers also report an uptick in sales of larger-screen TVs, he says: ?It?s beyond the cocooning we saw in the Jimmy Carter years.?

Sales of Sony?s first Ultra HD 4K TV, a $25,000 84-inch model that it began shipping in early December, have been ?exceeding expectations,? says Sony Electronics Vice President Brian Siegel. ?Over the last few years, consumers have been spending more time at home, and their expectations are increasing? for TV quality, he says.

Not so super socially
Super-cocooning is making us less social, says analyst Michael Greeson of The Diffusion Group, a media research group.

Technology makes it possible for us to avoid leaving our homes ? whether seeing a movie or getting food delivered ? and, he says, it can lessen our connections with others.

?With all the information and entertainment at arm?s reach at home, why get out and meet up with a friend when you can chat on Facebook? Why go shopping for a book at Barnes & Noble when you can search through a virtually unlimited bookstore like Amazon and never leave your couch??

Trend analyst Popcorn doesn?t see an end in sight for super-cocooning.

If anything, we will line our cocoons with more technology like the IllumiRoom that Microsoft showed off at the Consumer Electronics Show last month. Using a Kinect camera controller and projector, the IllumiRoom turns your entire room into a 3-D movie or game environment.

?You can see the evolution,? she says. ?But it all comes out of the same thing: We?re people getting more and more nervous about being vulnerable.?

Head and neck cancer molecular tumor subtypes documented

Feb. 22, 2013 ? Head and neck squamous cell carcinoma (HNSCC) is the seventh most common form of cancer in the United States, but other than an association with the human papillomavirus, no validated molecular profile of the disease has been established. By analyzing data from DNA microarrays, a UNC-led team has completed a study that confirms the presence of four molecular classes of the disease and extends previous results by suggesting that there may be an underlying connection between the molecular classes and observed genomic events, some of which affect known cancer genes. The clinical relevance of the classes and certain genomic events was demonstrated, thus paving the way for further studies and possible targeted therapies.

The study was published in the Feb. 22, 2013 issue of the PLOS ONE.

Neil Hayes, MD, MPH, associate professor of medicine and senior author, says, "Cancer is a disease caused by alteration in the DNA and RNA molecules of tumors. A cancer results when broken molecules initiate a cascade of abnormal signals that ultimately results in abnormal growth and spread of tissues that should be under tight control within the body.

"However, most common tumors, including head and neck cancer, have relatively little information in the public record as to how these signals coordinate to create different patterns of abnormalities. This study is among the largest ever published to document reproducible molecular tumor subtypes. Subtypes, such as those we describe, represent attractive models to understand and attack cancers for treatment and prognosis."

Dr. Hayes is a member of UNC Lineberger Comprehensive Cancer Center and national co-chair of the Data Analysis Sub-Group for The Cancer Genome Atlas, a program of the National Institutes of Health.

The team, composed of investigators from UNC and five other institutions, analyzed a set of nearly 140 HNSCC samples. By searching for recurrent patterns known as gene expression signatures, they were able to detect four gene expression subtypes. The subtypes are termed basal, mesenchymal, atypical, and classical based on similarities to established gene expression subtypes in other tumor types and expression patterns of specific genes.

In spite of being the seventh most common form of cancer in the United States, HNSCC is relatively under-studied in comparison to other tumor types, e.g. breast and lung. By leveraging the similarities found in the gene expression subtypes, the results of this study provide a connection to a range of well-established findings and additional insight into the disease.

Dr. Ben Carson on ABC's "This Week"

It?s a forum that isn?t known for making news ? so Dr. Ben Carson says the buzz created by his speech at last week?s National Prayer Breakfast,?including a Wall Street Journal staff editorial with the headline ?Ben Carson For President? ? came as a surprise.

?I don?t think it was particularly political,? Carson, the director of pediatric neurosurgery at Johns Hopkins, told ABC?s Chief White House Correspondent Jonathan Karl during an interview for??This Week.???You know, I?m a physician. I like to diagnose things. And, you know, I?ve diagnosed some pretty, pretty significant issues that I think a lot of people resonate with.?

With an audience that included President Obama and Vice President Joe Biden, Carson spoke out about political correctness, health care and taxes at the breakfast. In his roughly 23-minute address, Carson called for a private health care savings plan and a flat tax for all Americans. His address has since gone viral, racking up more than two million views on YouTube.

The speech isn?t Carson?s first foray into national politics. In 2008, President George W. Bush awarded the successful brain surgeon?the Presidential Medal of Freedom, the nation?s highest civilian honor.

On Sunday, Carson criticized some of President Obama?s policies but also called him a ?very talented politician.?

?There are a number of policies that I don?t believe lead to the growth of our nation and don?t lead to the elevation of our nation,? Carson added. ?I don?t want to sit here and say all of his policies are bad.?

He also offered his diagnosis for today?s political environment.

?What I would like to see more often in this nation is an open and intelligent conversation, not people just casting aspersions at each other,? Carson said on ?This Week.? ?I mean, it?s unbelievable to me the way people act like third graders. And if somebody doesn?t agree with them, they?re this and they?re that and, you know ? it comes from both sides. And it?s just so infantile.?

As for his own ambitions, Carson, 61, is retiring from practicing medicine this summer. But he told Jonathan Karl he?s not ruling out a future bid for political office.

?That?s not my intention,? he said. ?But I always say, ?I?ll leave that up to God.??

Israel?s Oscar-Nominated Film Prestigious Award

For the first time ever, an Israeli film has received the ?Cinema for Peace? prize from the Berlin International Film Festival.

The prize-winning documentary film, ?Gatekeepers?, which is also up for an Academy Award on February 24th, features some eye-opening, compelling and controversial conversations with a number of chiefs of Israel?s Shin Bet security agency, and is the first Israeli recipient of the prestigious award since it was first handed out in 2001.

The film, directed by Dror Moreh, won the Festival?s Best Documentary award in a ceremony at Berlin's Waldorf Astoria Hotel held by the Germany-based Cinema for Peace Foundation, a non-profit which supports films that deal with humanitarian and environmental topics.

The Cinema for Peace Foundation, which is made up of filmmakers and peace activists, was created after the 9/11 terror attacks in New York to shine a spotlight on humanitarian issues and human rights violations through film initiatives. This year?s jury is chaired by the Dalia Lama, and, in the past, Sean Penn and Robert De Niro have acted as Chair, along with many other popular actors.

SARS-linked virus may have spread between people

LONDON (AP) ? British officials say a mysterious virus related to SARS may have spread between humans, as they confirmed the 11th case worldwide of the new coronavirus in a patient who they say probably caught it from a family member.

The new virus was first identified last year in the Middle East and the 10 people who have previously been infected had all traveled there. According to Britain's Health Protection Agency, the latest patient is a U.K. resident with no recent travel to the Middle East but who had close personal contact with an earlier case. The patient is in intensive care at a Birmingham hospital.

The agency said this case was "strong evidence" for human-to-human transmission but insisted the risk to the general public was still considered very low.

Study in mice yields Angelman advance

Feb. 13, 2013 ? In the journal PLoS Biology, a team of scientists reports experiments showing how the gene defect of Angelman syndrome disrupts neurological processes that may be needed for memory and learning. In tests in mice, the team showed that a novel compound could restore the healthy processes.

In a new study in mice, a scientific collaboration centered at Brown University lays out in unprecedented detail a neurological signaling breakdown in Angelman syndrome, a disorder that affects thousands of children each year, characterized by developmental delay, seizures, and other problems. With the new understanding, the team demonstrated how a synthesized, peptide-like compound called CN2097 works to restore neural functions impaired by the disease.

"I think we are really beginning to understand what's going wrong. That's what's very exciting," said John Marshall, professor of medical science in the Department of Molecular Pharmacology, Physiology, and Biotechnology and the senior author of the study in the journal PLoS Biology. However Marshall did caution that it is too early to predict how soon a clinical therapy might arise from the results.

In mice and people, Angelman syndrome arises from flaws in a gene called Ube3A. When it functions properly, the gene limits the amount of a protein called Arc in the brain. Left unchecked by the disease, Arc impairs the development of synapses in the hippocampus. Those neural connections may be essential for proper learning and memory function.

In the new study, Marshall and his colleagues report a series of experiments that show how the abundance of Arc creates such negative effects and how Arc might possibly be defeated and its ill-effects repaired in the lab.

Essentially, Arc interferes with the operation of a synaptic protein called PSD-95, that is required for the actions of a growth factor, known as brain-derived neurotrophic factor (BDNF). This growth factor is released at synaptic contacts and initiates a sequence of molecular interactions necessary for the strengthening of neuronal connections or synapses. In mice with the flawed Ube3A gene, the signals sent by BDNF for memory formation are disrupted.

A team with a history

Although the researchers were surprised by the details they discovered about how Arc hinders the signaling process, they didn't come to the insight with complete naivete.

In other work, Marshall had been studying CN2097, designed by co-author Mark Spaller of Dartmouth College (Spaller synthesized it during earlier tenures at Wayne State University and Brown). The compound, which binds to PSD-95 was predicted to protect neurons under conditions of stroke and in disease states such as multiple sclerosis. With co-senior author Dennis Goebel of Wayne State, Marshall and Spaller found this to be the case. During the course of those studies the Marshall group learned that CN2097 enhanced the action of brain-derived neurotrophic factor (BDNF) which is known to be critically involved in long-term potentiation (LTP), a phenomenon believed by many neuroscientists to underlie learning.

Then, when University of North Carolina professor and former Brown postdoc Ben Philpot, now a leading expert on Angelman syndrome, returned to campus in 2008 to speak about Angelman syndrome, he showed how LTP is notably lacking in mice with the condition. After discussions with Philpot, Marshall and his group decided to test whether CN2097 might restore LTP in Angelman mice.

Early on, Marshall said, the team figured the defect in LTP in Angelman mice effect had to do with BDNF signaling.

"We started studying BDNF signaling in the Angelman syndrome mouse and discovered the signaling was defective, so that really was the breakthrough," he said. "It was completely unexpected. It's a new way of thinking about this disease."

That led to the set of experiments now reported in PLoS Biology, in which the group found that CN2097 essentially protects PSD-95 from interference by Arc, helping to restore BDNF signaling and the formation of stronger synapses. In electrophysiological tests on hippocampal tissue of healthy and Angelman mice, the compound made obtaining LTP significantly easier, although observing LTP in Angelman mice is still more difficult than in normal mice, Marshall said.

Paths to a therapy

Although the bottom line of the study is that the scientists now have taken a step toward understanding what seems to be going wrong in Angelman syndrome, the path toward a therapy for people is not so simple, Marshall said. For one thing, the team hasn't yet shown that mice benefit cognitively or behaviorally from CN2097's ability to protect PSD-95 from Arc and therefore to restore BDNF signaling and synapse formation.

"Can we actually rescue learning deficits?" Marshall said. "That would be the next stage to test. We haven't gotten that far yet."

Also, CN2097 breaks down in a matter of hours, so it may have to be administered quite frequently to sustain its beneficial effects.

But the research is extremely encouraging, Marshall said, because it pinpoints a potentially successful therapeutic strategy. Now scientists have two promising options. One is simply to keep testing CN2097 to determine whether it benefits live, behaving mice and what the best dosing regimen would be. The other is for scientists to take all that they have learned over the course of their experiments and use that knowledge to identify other compounds that might work like CN2097, but do so for longer periods of time.

"We think we are on the right track," Marshall said. "The goal is to try to help these kids. We want to do that as rapidly as possible."

Marshall has launched a new start-up company, Angelus Therapeutics, in hopes of raising the money required to perform more preclinical studies in animals. Further tests of the compound are necessary to lay the groundwork for possible future clinical trials.

Farther into the future, he added, the team can also look at other brain disorders that might also have to do with BDNF, synapse formation, learning, and memory, perhaps including Alzheimer's disease.

In addition to Marshall, Spaller, and Goebel, other authors are Cong Cao, Mengia Rioult-Pedotti and Crystal Yu at Brown; Paolo Migani at the University Pollitechnica delle Marche in Ancona, Italy; and Rakesh Tiwari and Keykavous Parang at the University of Rhode Island.

The Angelman Syndrome Foundation, the Rhode Island Science and Technology Advisory Council, and the National Institutes of Health (Grant R21-NS061176) supported the research.
